****![herzog_edit](https://bpi.bioprocessintl.com/hs-fs/hubfs/ATE/mohammed-sm.png?width=133&name=mohammed-sm.png)****

**Dr. Adil Mohammaed**

Staff Fellow, US FDA

Dr. Mohammad received his Ph.D. in Chemistry from The University of Iowa in 2010 and joined FDA and as a Post-Doctoral Scholar in 2012 in the Division of Product Quality Research (DPQR) at CDER (Center for Drugs). His regulatory research efforts have focused on physiochemical characterization of Prussian blue (a medical countermeasure drug). His research also included development and validation of analytical methods using HPLC, UPLC and ICP-MS to support product quality issues in drug substance and drug product formulations. His most current research is focused on developing and validating analytical methods to screen and quantify trace metals in commonly used bioreactor media. He has also done comprehensive research on studying the effect of trace metals on the production and quality of therapeutic proteins.

**![nagendra_edit](https://bpi.bioprocessintl.com/hs-fs/hubfs/ATE/Madhavarao-sm.png?width=133&name=Madhavarao-sm.png)**

**Dr. Chikkathur Madhavarao**

Biologist, US FDA

Dr. Chikkathur N. Madhavarao obtained doctoral degree in Biotechnology from University of Pune, India with a focus on molecular interactions in biological macromolecules and bioenergetics. Later he pursued post-doctoral research on congenital rare diseases that occur due to deficient or mutant enzymes and normally affect the nervous system. After joining FDA in 2009 he pursued to understand the impact of production processes on product quality attributes of a model for enzyme replacement therapy (ERT) class of therapeutics. Variation in the metal concentration in the cell culture medium during production and its impact on product quality attributes (PQAs) of the therapeutic proteins is one of the research areas of interest of Dr. Madhavarao. He is currently focusing on improving the manufacturing efficiency of protein therapeutics to help produce products with homogeneous PQAs.

In addition, Rob has authored 5 textbooks on the fundamental principles and applications of ICP-MS. Rob has an advanced degree in analytical chemistry from the University of Wales, UK, and is also a Fellow of the Royal Society of Chemistry (FRSC) and a Chartered Chemist (CChem).

## About this event:

#### Date: December 2, 2021

#### Time: 11 am EDT | 8 am PDT | 5 pm CEST

#### Duration: 45 minutes

Biotechnology products (biologics) are often produced from mammalian cells grown in large-scale bioreactors. The dynamic environment within the bioreactor is comprised of growth media along with cells, proteins, nutrients of organic and inorganic origin, metabolic waste products and metal ions. Metal ions can act as enzyme cofactors and can directly influence the kinetics of biochemical reactions that may also influence the biological production and quality attributes of therapeutic proteins. As a result, variations of the metal content in the cell culture media can affect the cell growth and production of the biologic product as well as its critical quality attributes (CQAs). The webcast will focus on the use of inductively coupled plasma mass spectrometry (ICP-MS) to study the role of metals in the production of biologics.

The first two speakers will describe an ICP-MS analytical method to determine the composition of a panel of metals in different cell culture matrices, which was then used to validate and screen cell culture media, determine changes in metal concentration during cell growth, evaluate the effect of spikes in metal concentration, and study the impact of zinc supplementation on purity and yield.

The second speaker will focus on the benefits of a novel multi-quadrupole ICP-MS system to mitigate common polyatomic spectral interferences found in cell culture media samples. By utilizing a true quadrupole with dynamic bandpass tuning (DBT) in the collision/reaction cell, it allows the use of any reactive gas to either move the interference away from the analyte mass using MS/MS mode or shift the analyte mass from the interference using Mass Shift mode. The mechanism of interference removal will be demonstrated using examples of metals commonly monitored in cell culture media.

Three takeaways from this webcast:

1. Complexity of cell culture media with large variations in metal concentrations
2. Trace-element analysis of cell culture media poses challenges for conventional ICP-MS
3. Multi-quadrupole ICP-MS offers unique interference removal capabilities, improving accuracy and lowering detection limits
