| Monoclonal antibody (mAb) manufacturing continues to reach new levels of productivity, with upstream titers of 5–10 g/L becoming increasingly common. Standard downstream purification schemes remain an opportunity for further efficiency gains. The reduction of product-related impurities is traditionally handled by bind-and-elute cation exchange chromatography (CEX) unit operations, but can be limited in capacity and productivity. Flow-through hydrophobic interaction chromatography (HIC) can be an effective alternative, enabling required impurity clearance at much higher loadings, potentially delivering a more efficient, cost-effective process. This webinar will provide a start-to-finish overview of the process development behind transforming a typical bind-and-elute (B/E) CEX step to a flow-through HIC operation, from evaluating POROS HIC resins to scaling conditions for pilot runs to support consistent performance during tech transfer to manufacturing. Key Takeaways: - Flow-through HIC enables significantly higher product loadings compared to traditional bind-and-elute CEX, improving process efficiency and cost-effectiveness. - HIC delivers required product-related impurity removal while accommodating increasing upstream titers of 5–10 g/L in mAb manufacturing. - The complete process development workflow from resin evaluation through pilot-scale optimization allows successful tech transfer and manufacturing implementation. |
