Trehalose Enhances Removal
of Host Cell-derived Impurities During
Harvesting Process of Antibody Production

Advancements in upstream processing for antibody production have increased host cell-derived impurities, posing challenges for downstream purification. To address this, cell line and antibody engineering and clarification devices have been explored. This study investigates trehalose, a non-reducing disaccharide known for cytoprotective effects, added after CHO cell cultivation. Trehalose reduced host cell DNA and proteins during harvesting with diatomaceous earth and 3M™ Harvest RC. It also lowered conductivity, enhancing impurity clearance. These effects persisted through Protein A chromatography. Trehalose presents a novel strategy to improve purification efficiency in therapeutic antibody manufacturing. Key Takeaways:
